The Interaction of Carcinogenic Hydrocarbons with Tissues VI. Studies on Zero-Time Binding to Proteins*
نویسنده
چکیده
A comparison has been made of the methods used for homogenizing and processing tissues in hydrocarbon-skin protein binding studies. The procedure of choice is to use a glass homogenizer cooled in ice and to precipitate the proteins with acetone. The zerotime binding of a number of hydrocarbons has been studied and has been found to be negligible. The zero-time binding of several derivatives of 1,~,5,6-dibenzanthracene has been studied, and in general they bind much more than the parent hydrocarbon, especially ~-phenylphenanthrene-3,~'-dialdehyde, which is not carcinogenic. An at tempt was made to find out whether this compound is a metabolite of 1,~,5,6-dibenzanthracene in mouse skin in vivo. Only negligible amounts could be detected. The significance of these results for the hypothesis that the formation of a tissue-carcinogen complex is an early step in carcinogenesis is discussed.

Studies were carried out in which the binding of thirteen carcinogenic and noncarcinogenic hydrocarbons to the soluble proteins of mouse skin was compared. A good quantitative correlation was found to exist between the binding of the hydrocarbons to a particular protein fraction (protein fraction 1) obtained from mouse skin, which migrates toward the cathode with a relative mobility of +0.23, i...

Studies have been carried out on the nature of the proteins of mouse skin to which hydrocarbons are bound after topical application. No relation was found between the carcinogenic activities of various hydrocarbons and their inhibition of the incorporation of DL-lysine-2-C 1~ into mouse skin proteins. Salt fractionation of the proteins to which 1,~,5,6-dibenzanthracene-9,10-C TM is bound reveal...

Human serum albumin is one of the most important blood proteins that has the ability to bind a wide range of compounds and different drugs. Hence, knowing how drugs bind to albumin is crucial to understand their pharmacokinetics and pharmacodynamic properties. The binding of drugs to protein affects the drug's excretion, distribution and interaction in the target tissues. Nicotinamide (NA) is a...
